Reading from a few posts above by ktula. It sounds like fw 3.0 did not include a baseband upgrade. Howver, you will loose your activation & jailbreak.
In this case, a DFU restore to 2.2.1 + quickpwn should bring you back to life.
This is only a guess since I don't have 3.0.

Unfortunately, i have this problem like many others. quickpwn does not recognize my iPhone.
This is how i got myself into this "mess":
- Installed iPhone 3.0 beta (for iPhone EDGE) on my iPhone EDGE (or original iPhone)
- iTunes/iPhone was whining that my SIM was not from an supported carrier. Did not realize i needed the AT&T SIM. So i removed my T-Mobile SIM and inserted an AT&T SIM that i borrowed.
- That got past the error but now because my iphone is not yet registered on the developer site as a test device, i was prompted to do so. Since iTunes did not recognize my iPhone, i had to determine the UDID by running xcode from iPhone 3.0 beta SDK.
- Once i entered the UDID on the developer site, i was able to get 3.0 running on my iPhone.
- I tried to replace the SIM with my T-Mobile SIM but that caused iPhone to be placed in emergency call mode.
- Removing the SIM allows all functionality (except phone) of iPhone.
- I tried to restore the stock 2.2.1 firmware. The restore appeared to work but iphone rebooted, the picture showing a USB cable connecting to iTunes appeared. With a T-Mobile SIM, iTunes (8.1) complained that the SIM is not valid. Quickpwn did not even see the iPhone.
- I also tried to restore to a 2.2.1 firmware created by pwnagetool. Unfortunately, the end result is the same as the stock 2.2.1 firmware.
So am i SOL now?

Unlock and activation remains with update method
I used an pwned 2.2.1 on my 1G iPhone and upgraded to 3.0 and my phone remained activated and it works fine and syncs with iTunes (I do however use an O2 sim (official carrier).
Also note I removed my O2 sim and put in a T-Mobile one and the phone still works, so the unlock is there too.
Not sure about this whole deal with needing dev account? Maybe for 3G phones?
Cheers.
Note: 02 is the official carrier in the UK, so the unlock appears to still be there!

@ktula: Try putting it in DFU mode before connecting it to Quickpwn.
Can someone PLEASE run a MD5 hash check on their 2G 3.0 firmware and post it here?

My Unlock+Activation stayed.
I did:
just an upgrade to 3.0 (Shift click on upgrade)
I have:
A paid developer account
an iPhone 1G US
i live in Luxembourg (i think all carrier are allowed here)
I won't talk about the beta as it is under NDA, but it is goood

I finally managed to use Quickpwn to activate/jailbreak my iphone, which had been downgraded from 3.0 to 2.2.1. However, i had to run Quickpwn on a windows xp computer, not my Mac. I still can't get Quickpwn on my Mac to recognize my iPhone. May be it has something to do with this iTune message that i have been getting when i connect iPhone to my Mac:
iTunes was unable to load dataclass information from
Sync Services. Reconnect or try again later.

Did you do your upgrade to 3.0 beta from a Mac or a windows-based PC?

May be the question that i should be asking is: where did you get your 3.0 beta? I downloaded mine from Apple's iphone developer site.
