This has been answered many many times.
Please look around the forum ...
the short answer is your phone is pretty screwed at this point.
you have to wait for new version of pwnagetool/QuickPwn AND yellowsn0w !!!

Wow after all the warnings about not updating to 2.2.1 you had to go do it!!. well my friend you are well and truly shafted at this moment in time. there is currently no solution to reverse your baseband and the dev team have said that the update may stop the unlock forever if you update.

Looks like you have a paper weight for your desk

i read on 2.2 about people just using update and it stays unlocked but this was nto fthe case for me on 2.2 it re locked but luckily the same day there was quickpwn... no need to rush and upgrade, may even skip this upgrade and wait for 2.3 where bluetooth will connect to itunes? or is that lies

Regardless of 2.2, 2.2.1 or 2.3 .... If there is a change in the Baseband, you are completely screwed (not for the 2G though). Remember that DevTeam constantly warn people not to update.

For iPhone 3G, yellowsn0w is "software" proxy that runs on the iPhone. It is not a complete unlock like iPhone 2G. The new baseband on iPhone 3G may close certain things that prevent yellowsn0w from working; therefore newer version of yellowsn0w may be not out for a while.

The only way to update to newer OS that runs on the application processor is to use pwnageTool to build your custom image WITHOUT the baseband upgrade option. With that said, every new firmware will require a pwnagetool update.

    Default give it a try

    Hi!

Maybe you want to give it a try and reinstall yellowSn0w running 2.2.1.
Therefor you first have to jailbreak your phone!
The 'other' DEV-Team just released their jailbreak. (atm MAC only)
    hXXp://ripdev.com/pusher (substitude T for X )
IMHO it unfortunately 'only' installs Installer.app
You have to add this source to install yellowsn0w:
    hXXp://i.yellowsn0w.com/

Even if yellowsn0w does not work, your iPhone is now jailbroken.

Yellowsn0w won't work, it has already been confirmed that the hole that YS used has been patched.

isn't it strange that some people who have gone to all the trouble to find out how to jailbreak and unlock their phones still fail to see/read the warnings about updating FW until safe to do so, this has been a known thing since when the first unlocks came out for the 2G phone. Also isn't it funny how many people have problems messing with their "freinds" phones but not their own. (slightly off topic i know)
