    Sorry, but they are gone. I know there are forensic methods trying to recover data from an iPhone. But this is way tooo much trouble and there is no guaratie that this will bring back any photos.
    even if you dont have any music or video on your iPhone it makes sense to sync, you do have another backup of photos, addresses, sms- and call-history and stuff.

    What is the best app/method to back-up the contents of the iPhone?

    Well iTunes does a good job, it backup the stuff once you sync. You could - on a jailbroken iPhone - use OpenSSH and SFTP the files inside
    /private/var/mobile/Library
    it's all in there.
    But as I said, iTunes does a pretty good job, I have used the backups several times, restored the data from it and put it back to my iPhones. Due to testing a lot I often have to DFU restore and dont use the iTunes backup, but just the few data from the above path.
    depends on what you do to your iPhone ;-)

    Thanks. Also is it ok to re-do the 2.2.1 upgrade (even if phone is already at 2.2.1? I ask because I carried out a restore of backup that was done before the upgrade and after the restore my apps (from app store) suddenly did not work and had to reinstall them. But I don't know what else the restore has broken so I was thinking of doing fresh 2.2.1 install. Would I just use the same method to install 2.2.1 again and is there anything special I would need to do since the phone is already at 2.2.1 or just follow that amazing iclarified guide?

    You can de-do the 2.2.1 restore anytime again. If you want to get rid of all old data DFU restore to 2.2.1. After that follow the iclarified tutorial to jailbreak/activate/unlock.
    First time then connecting to iTunes, the programm will offer to recover data from an old backup. This is where I always choose "setup as a new iphone". Then I choose my sync settings in itunes, done.
    Restoring from the backup is nice and handy but: it will not only bring back some data like sms- and caloing history but also old preferences.
    If you for example had been on a 1.x firmware those settings might not work at fw 2.x. Or if prefs get corrupted and you bring em back, you're stuck again.
