I see that there is only the sync mode available with the iPhone,
does anybody knows a way to fill the iPhone manually with iTunes ?

The short answer is no. The iPhone has adopted a new protocol similar to that used on the Apple TV, and at this point there is no manual mode via iTunes itself.
Third-party tools and hacks into the iPhone's file system may eventually provide this capability outside of iTunes (ie, using third-party software), but at this point iTunes does not offer this capability directly. A future update might change this, of course.
The closest thing you can do right now with iTunes is to sync a selected playlist, and then just manually put your tracks into that playlist. This is almost as practical as a manual sync unless you're loading music on from more than one computer, or you don't want to keep your music on your computer.

I found out by accident that if you 'fake activate' the iphone and don't go through the AT&T screen in iTunes, you can just drag and drop videos/music onto the iPhone but you don't get any other syncronisation (photos, contacts, calendar, etc).... Once you properly register the iphone in iTunes this abilty goes away. I just create a playlist called iPhone in iTunes which I use to sync now.
