Yes, you will need to generate a custom firmware with pwnagetool for keeping the unlock active. If you are not using an official Apple carrier, remember to select the "Activate the phone" option. After generating the firmware, you will have to restore it with iTunes (Option + Restore in Mac, Shift + restore in Windows). Then, select your custom firmware.
After the restore (and after restoring your latest backup), you will have a jailbroken iphone, but no unlocked. To unlock it, you need Cydia. Add the repo repo666.ultran0w.com to the cydia sources, then install UltraSn0w from that source and reboot. Remember: You will need WIFI to install UltraSn0w.
After that, your iphone will be upgraded, jailbroken, unlocked and activated
